Anti-inflammatory and immune enhancing activities of PB203 in mouse macrophage RAW264.7 cells

Macrophages secrete various cytokines and inflammatory mediators, resulting in playing critical roles inflammation immunity. In this study, we investigated anti-inflammatory immune enhancing properties of PB203, which is a water-soluble extract powder from the fruit Actinidia polygama, macrophages. A. polygama medicinal plant traditionally known to treat abdominal pain, stroke rheumatoid arthritis. However, molecular mechanism for modulation PB203 still unclear. Therefore, assessed effects on lipopolysaccharide (LPS)-induced activation, elucidated its action mouse macrophage, RAW264.7 cells. significantly suppressed not only levels nitric oxide (NO), prostaglandin E2, tumor necrosis factor-α (TNF-α) interleukin-1β (IL-1β), but also mRNA expression inducible NO synthase, cyclooxygenase-2, TNF-α IL-1β LPS-stimulated We found that these activities were mediated through inhibition toll-like receptor 4 nuclear factor kappa B (NF-κB) induced by LPS. On other hand, normal macrophages, dose-dependently elevated gene immunomodulators including granulocyte-macrophage colony-stimulating factor, granulocyte monocyte chemoattractant protein-1 statistically significant manner. The IL-10, IL-1β, IL-6, interferon-γ remarkably upregulated treatment 500 μg/mL PB203. addition, PB203-mediated production was attenuated NF-κB Interestingly, promoted erythroid-2-related 2, increased level heme oxygenase-1, representative antioxidant enzyme, both Taken all together, results suggest may have great potential as candidate agent improving diseases or preventing infectious diseases.
